The fifth seeds outclassed veterans Rohan Bopanna and Ivan Dodig in the Montreal Masters doubles final 6-4 3-6 10-6.
Indian Bopanna and his partner Dodig saved two break points in the ninth game of the second set to make it all tied at one set apiece.
Herbert, 26, and 35-year-old Mahut got off to an early 4-0 super tie-break lead and never looked back to seal the title.
The French pair won 88 per cent of their first serve points in an hour and 21 minutes spent on the court.
Back in May, Herbert and Mahut won it all on the clay of the Rome Masters.
Herbert and Mahut, the 2015 US Open and 2016 Wimbledon champions, have yet to win a Major this year. The duo will be hoping to continue their good form at the Cincinnati Masters before they head to Flushing Meadows.
Dodig and Bopanna, who were playing just their second tournament as a pair, fell just short of landing a Masters 1000 title.
Fifth-seeded Herbert and Mahut, as well as seventh seeds Bopanna and Dodig, have a bye round one at Cincinnati's Western & Southern Open.
