Fan-favorite Isner outclassed world No. 45 Viktor Troicki 7-6 (3) 6-4 to open up his Cincinnati Masters campaign on a good tale.
Serb Troicki saved a set point in the 12th game to clinch a tie-break, where he stood no chance against the American.
The 2013 runner-up then held on to his serve after breaking Troicki in the opening game of the second set.
Isner was dominant on his serve, firing 20 aces and winning 91 per cent of his first serve points in an hour and 25 minutes of play.
He will be meeting next an American player as he awaits the winner of the match between Donald Young and Tommy Paul.
Experienced Muller found a way to earn a narrow 6-4 5-7 7-6 (3) win over unseeded local Ryan Harrison.
The 16th seed saved four break points during the third set before clinching the win.
The American had a 2-1 tie-break lead when Muller went on to win five points in a row.
Muller hit 21 ace shots and won 90 per cent of the points played behind his first serve. He also saved 5-of-6 of the break point tries he faced in just over two hours of action.
Next up for him will be either Monte Carlo Masters runner-up Albert Ramos or qualifier Mikhail Youzhny.
